In the current version of the protein production pipeline of the Joint Center for Structural Genomics (JCSG, www.jcsg,org), all protein targets up to a molecular weight of about 50,000 are screened for suitability of the particular construct for crystallization trials and structure determination either by X-ray crystallography or by NMR spectroscopy. This lecture will survey the experience gained with the use of NMR screening in structural genomics, and discuss the role of NMR for de novo three-dimensional protein structure determination in solution.
