An automated advance over high resolution screening is described that provides complementary metabolomic structural information with on-line fraction collection. Nanoelectrospray infusion (nanoESI) of fractions from commonly used LC columns with a post-column splitter to simultaneously acquire LC/MS/MS data and fraction collection once every 30 seconds on a single platform is discussed. We compared both LC/MS and chip-based infusion nanoESI of LC fractions containing ibuprofen metabolites in urine. Thirty percent more metabolites were identified over traditional LC/MS/MS analysis. Furthermore, this accelerated sample screening of any complex matrix via multi-channel analysis includes the benefits offered by nanoESI analysis including: enhanced dynamic range on-line, extended analysis times that permit data averaging, collision energy optimization and time to perform subsequent scan types such as MSn, and precursor or neutral loss scanning. Furthermore, the technique allows for sample portability or immediate sample re-scrutiny, P450 affinity or drug-target interaction screening.
