The development of robust functional assays and of useful devices for screening extensive collections of compounds on ion channels, has been particularly challenging so far. Therefore the creation of cell based assays tailored for the use on different technologies represents a valuable tool for pharmaceutical companies. In this work we have cloned and stably expressed in mammalian cells, several different ion channels and transporters. Functional cell-based assays suitable for use in HTS have been developed and validated using both fluorescence method and the classic patch clamp technology. A comparison of the data obtained with different technologies (FLIPR384, standard patch clamp, patch clamp robot Flyscreen 8500, luminescent measurement) is made and commented for several different ion channels, with a particular emphasis on the HTS application. |
