Mostacero, Manolo Valmaseda and Julio Martin, GlaxoSmithKline Presenting Author: Laura Vela, GlaxoSmithKline - Spain HTS assays configured in such a way that allow multiple readouts from a single testing well offer clear advantageous over single readout formats. In particular, assays running in continuous kinetic mode may exhibit higher signal-to-noise window, higher robustness to interferences, in-well quality control for hit (in) validation and high information content valuable for mode of action. We present the case of an uHTS campaign of an enzyme using a fluorogenic substrate. Data analysis has been fully automated for the calculation of enzyme kinetics parameters as estimators of the activity and reliability of compound response. |
