High Content Screening is multiplexed, functional cell-based screening. At Serono Pharmaceutical Research Institute in Geneva HCS has been used in the past for secondary screening of small molecules as well as in signaling and toxicology research. Today, we use HCS in primary screening for therapeutic protein and small molecule discovery. At Serono, we have applied HCS technology early on in the screening process to eliminate toxic compounds, and select the best positives. The application of HCS in primary cell-based screening has enabled us to select positives with a greater chemical diversity. At the same time, we use an HCS-based genotoxic assay measuring micronuclei formation in mammalian cells, to eliminate genotoxic compounds. Together, this strategy has enabled us to select higher quality positives, optimizing the chances of small molecules progression. |
