The need for more specific information on a molecular level for the detection of metabolic and genetic changes in vivo has grown since novel molecular imaging technologies have been established in drug discovery.
We have combined high resolution fluorescence imaging with the photon statistics based single molecule detection to analyze single living cells. Using the same positioning and detection system for laser scanning based imaging and point fluctuation acquisition, accurate spatial relation can be achieved with conserving single molecule information for quantitative determination of concentrations.
We will present first measurements of calcium release and membrane potential detection in adherent CHO cells using the advantages of image processing and photon statistics methods FIDA and FIDA-2D. Using dual color coincidence information evaluated by fluctuation analysis a total intensity independent model can be applied for characterizing cellular inhomogeneous samples.
