It is useful for functional assays, binding assays requiring protein immobilization and purification. SNAP-tag can be labeled in living cells with cell permeable fluorescent substrates, even for two color pulse-chase protocols. SNAP-tag can also be labeled with affinity tags like biotin. SNAP-tag and its labels withstand typical fixation protocols, allowing easy combination with antibody labeling as well as the use of a wide range of non cell-permeable SNAP-tag substrates on fixed cells. Surfaces coated with SNAP-tag substrates allow selective immobilization of SNAP-tag fusion proteins from complex samples. One immobilization application is pull down assays on substrate coated beads. The combination of labeling applications - in solution, inside or outside of live or fixed cells - with specific immobilization makes the SNAP-tag the first true multipurpose protein tag. Last not least, the implicit licensing strategy makes for easy access to this versatile technology. |
