Simultaneous Image Analysis of Multiple Apoptotic Pathways

    Apoptosis, or programmed cell death, is involved in almost every physiological and pathological process. There are numerous processes involved in apoptosis, some in sequence and others simultaneously. Using image analysis modules of MetaXpress™ we examined three separate parts of the apoptotic pathway to determine whether the responses would be differentially sensitive to the toxic effects of staurosporine treatment. For changes in the mitochondria we looked at translocation of Cytochrome C, AIF and Bax to the mitochondria. For disruption of the nucleus we used nuclear morphology and for cytoskeletal changes we used cell morphology. Although all treatments produced similar responses and IC50s there were subtle differences between the responses that might otherwise have been missed in a single parameter analysis. These results indicate that these assays may be multiplexed for finer discrimination of apoptotic pathways.
