A main trend in efforts on improving immunoassays is to reduce sample and reagent consumption and turnaround time for the assay. The Gyros CD is a lab-on-the-chip consisting of 96 individual structures molded in a plastic CD. 200 nL samples are driven through the structures by spinning CD, thus creating centrifugal force. Sample and reagent volumes are defined within the CD. It has been shown that the CD technology is an efficient way to totally automate sandwich immunoassays to produce clinical-quality analytical results. The analysis of low-molecular weight biomolecules calls for the competitive immunoassay format associated with the use of only one antibody. The Assay Designs Substance P immunoassay detects this important 12 amino acid peptide and has been modified to demonstrate the possibility to perform competitive immunoassays in the CD. We compare samples in the automated CD format and the traditional ELISA format assay.
